The must have Glyphs are:
20% more threat to Challenging Shout
100% more threat to Shield Counter
2x duration of Debilitate

The Good ones are
25% more damage to Shield Counter
50% stun duration of Shield Bash
25% cooldown reduction on Shield Bash

Everything else is up to you.

For crystals, threat crystals don't stack as it states on the tooltip. Fill the rest with either Mana regeneration crystals or boss damage crystals. For armor crystals use boss damage reduction, they can be quite expensive so using health increase is another choice. To level just use full health and mana regeneration crystals. Since you duo with a priest, you can swap the health regen to health boost. But soloing regeneration is amazing.
Yes all lancers attacks do reduced damage when dealing damage to more than 1 target. The positive thing though, is that threat moves such as Shield Counter is a set amount so it will not diminish when you are tanking a mob. However questing and grinding speed you are out of luck. All classes suffer AoE damage reduction with the exception of Penetrating Arrow and other pierces which works a bit differently as it does full damage to the first target then less for each target it passes through.
Shield Counter with the threat glyph and Challenging Shout are your main threat generating skills. Anything else does negligible threat. Try and use them as much as possible over your other skills and threat shouldn't usually be a problem. If you are still having problems threat crystals along with threat enchant on your weapon might help as well.
Blocking has to be the most satisfying skill. Nothing better than waiting until the last second to hit block and hearing the clang with a popup going Defense Successful!
Well, I'm 54 currently and I haven't been to the very end dungeons but I'll share some feedback. If you have 6 mobs on you theres rarely any chance that you can do any damage. If you unblock for even a split second you'd die instantly. Try and keep pulls to 2-3 unless its a pack of weaker enemies that comes in 6. If you find yourself losing aggro often, invest in some threat glyphs like Challenging Shout and Shield Counter threat. Weapons also have enchants with 13.5% more threat as well as crystals that give bonus threat. Those are unique so you only need to use 1 in your weapon.

Once you understand the pattern of mobs and bosses you can start being more aggressive. If you glyphed Debilitate the duration increases to 30 seconds giving you more time to use offensive skills before needing to restack it. I have ran with healers who doesn't heal me very often. In those cases its better to just survive. You have more than enough threat with Shield Counter and Challenging Shout. Your main job is to maintain threat any damage you do is bonus damage. If you feel unsafe as its your first run don't be afraid to block more. Killing it a bit slower is better than dying and wiping. Your DPS is almost completely insignificant compared to the DPS. Thats what they are here for, to kill the boss that you tank.

You shouldn't see Mystics with their tanking Thrall. If they do ask them nicely to not use it. I haven't really lost aggro to a Healer. Only people I lose aggro to are DPS when the boss chains back to back red glare moves.

I do have a video of me tanking http://www.youtube.com/watch?v=J7GSysjt-B4

I never edited anything to make me look better this is just how I usually tank. Its just me solo queuing and running a dungeon for the second time. It is a little old as I started to learn to tank a bit differently.
Shield Bash with glyphs can bring it down to 19 seconds cooldown with a 6 second duration stun. That is massive. Leash however has a 30 second cooldown and lasts around 1 second. Leash is also needed for bosses when they run to the center to summon or cast dangerous magic and should not be used every time it is off cooldown while Shield Bash can.

Mostly Leash's use in PVE is to stop BAM's moves or reposition pullable mobs. The threat on it is nothing compared to Shield Counter or Challenging Shout as you would be using them all the time to tank in a dungeon. I wouldn't recommend glyphing it in PVE.

In PvP I would still rather use Shield Bash. A Shield Bash into an Onslaught is a lot of pain. Leashing someone with the stun guarantees a Shield Bash stun. But nothing is stopping you from getting both in PVP.
Do not waste leash to interrupt a red glare move. Its 45 seconds or 30 seconds when rank 2. Not only that it uses a long cooldown if the stun doesn't go through you're stuck in the animation and get smashed for massive damage. If your leash is on cooldown while the boss runs to the center and summons a bunch of stuff you'll be unable to interrupt.

Personally I like to use Challenging Shout a lot as its your main threat skill while tanking. It might not be needed early on but as DPS damage increase and yours slow down you need every bit of extra threat you can push in.

Stack Debilitate and Shout whenever it is safe to do so while Shield Countering every cooldown is all you need to tank. Anything else you do is just bonus damage and to relief stress by stunning them.

I made a mini video on tanking. Its not very well edited or hard content but it should give you an idea on tanking.

edit. Oh also one thing to watch out for is temporary aggro loss with the half closed orange and red eyes. You will first meet stuff in GL at around lv 48 that will drop aggro. Don't panic that is normal you can't do anything about it. Some bosses will drop target on you too. As long as you see the orange circle around your feet it means you have maximum threat and is the focus target. They will return to you after the temporary aggro is lost. I can't count the number of DPS that scream at me when this happens.

I've noticed that the snake nagas can attack over me and hit me when i'm blocking in their face. I'm not sure if its because I'm an Elin lancer and really short or if all races suffer from that problem
Am I the only one that is disappointed that there isn't enough hair up armor? I miss my T2 with the side ponytail
